IFN-y Induces Cell Growth Inhibition by Fas-mediated Apoptosis: Requirement of STATI Protein for Up-Regulation of Fas and FasL Expression1

چکیده
The mechanism by which II Vy inhibits tumor cell growth has not been fully understood. Here we report that IFN-y up-regulated the ex pression of Fas and Fas ligand (FasL) on HT29 cells, a human colon adenocarcinoma cell line, and subsequently induced apoptosis of these cells. The kinetics of cell death in IFN-y-treated HT29 cells paralleled the increase in the levels of Fas and FasL expression. We further show that IFN-y up-regulated the expression of Fas and FasL in STATl-transfected U3A cells but not in STATI-deficient U3A cells. Correspondingly, IFN-y induced cell death in STATl-transfected U3A cells but not in STATIdeficient U3A cells. IFN-y-induced cell death was inhibited by caspase-1 inhibitors. Our results suggest that cell growth inhibition by IFN-y is due to apoptosis mediated by Fas and FasL interaction.

متن کاملCrosstalk between keratinocytes and T lymphocytes via Fas/Fas ligand interaction: modulation by cytokines.
Apoptosis mediated by Fas/FasL interaction plays an important role during many inflammatory skin disorders. To estimate whether the expression of FasL, the ligand for Fas, might be regulated by cytokines we stimulated primary human keratinocytes with several pro- and anti-inflammatory cytokines. Keratinocytes cultured to subconfluence expressed FasL constitutively. Cells stimulated with the pro...
متن کاملOxysterol-binding protein-related protein 8 (ORP8) increases sensitivity of hepatocellular carcinoma cells to Fas-mediated apoptosis.
Human hepatoma (HCC) has been reported to be strongly resistant to Fas-mediated apoptosis. However, the underlying mechanisms are poorly understood. In this study the function of oxysterol-binding protein-related protein 8 (ORP8) in human hepatoma cells apoptosis was assessed. We found that ORP8 is down-regulated, whereas miR-143, which controls ORP8 expression, is up-regulated in clinical HCC ...
متن کاملFas-mediated apoptosis in Ewing's sarcoma cell lines by metalloproteinase inhibitors.
BACKGROUND Fas ligand (FasL) is a transmembrane protein that induces apoptosis (programmed cell death) in susceptible cells by interacting with its receptor, Fas. Transmembrane FasL is cleaved by a metalloproteinase enzyme into a soluble form that is released into the extracellular medium. Tumors of the Ewing's sarcoma family express functional transmembrane FasL and release soluble FasL. This ...
متن کاملDifferential regulation and function of Fas expression on glial cells.
Fas/Apo-1 is a member of the TNF receptor superfamily that signals apoptotic cell death in susceptible target cells. Fas or Fas ligand (FasL)-deficient mice are relatively resistant to the induction of experimental allergic encephalomyelitis, implying the involvement of Fas/FasL in this disease process. We have examined the regulation and function of Fas expression in glial cells (astrocytes an...
